RESUMO

The efficient utilization of food waste (FW) resources through Food Waste Valorization (FWV) has received increasing attention in recent years. Various decision-making studies have been undertaken to facilitate FWV implementation, such as the studies on decision-making framework and FWV technology assessment. Food waste hierarchy is a widely discussed framework in FW management, but it was found too simplified and does not always contribute positively to environmental sustainability. Moreover, decision-making studies in FWV often focus on specific aspects of the food system and employ distinctive decision-making approaches, making it difficult to compare the results from different studies. Therefore, our literature review is conducted to provide a comprehensive understanding of FWV decision-making. This study identifies what decisions are needed, and three levels of decisions are revealed: system-level, FW stream-level, and FWV option-level. The assessment approaches and criteria used to support decision-making in FWV are also collected and analyzed. Building upon these findings, an hourglass model is synthesized to provide a holistic illustration of decision-making in FWV. This study untangles the complexities of FWV decision-making and sheds light on the limitations of current studies. We anticipate this study will make more people realize that FWV is a multidisciplinary issue and requires the collective participation of researchers, practitioners, policymakers, and consumers. Such collective engagement is essential to effectively address practical challenges and propel the transition of the current food system toward a more resource-efficient paradigm.

Global climate change has significantly impacted the production of various crops, particularly long-term fruit-bearing plants such as citrus. This study analyzed the fruit quality of 12 citrus orchards (Citrus Sinensis L.Osbeck cv. Bingtang) in a subtropical region in Yunnan, China from 2014 to 2022. The results indicated that high rainfall (>220 mm) and low cumulative temperature (<3150 °C) promoted increases in titratable acidity (>1.8 %) in young fruits. As the fruits further expanded (with a horizontal diameter increasing from 50 to 65 mm), excessive rainfall (300-400 mm), lower cumulative temperature (<2400 °C), and a reduced diurnal temperature range (<10 °C) hindered decreases in titratable acidity. Conversely, low rainfall (<220 mm), high cumulative temperature (>3150 °C), and a high diurnal temperature range (>14 °C) promoted the accumulation of soluble solids in young fruits (9 %) at 120 days after flowering (DAF). Furthermore, low rainfall (<100 mm) favored the accumulation of soluble solids (1.5 %) during fruit expansion (195-225DAF). To quantify the relationship between fruit acidity and climate variables at 120 DAF, we developed a regression model, which was further validated by actual measurements and accurately predicted fruit acidity in 2023. Our findings have the potential to assist citrus growers in optimizing cultivation techniques for the production of high-quality citrus under increasingly variable climatic conditions.

The exogenous application of amino acids (AAs) generally alleviates cadmium (Cd) toxicity in plants by altering their subcellular distribution. However, the physiological mechanisms underlying AA-mediated cell wall (CW) sequestration of Cd in Chinese cabbage remain unclear. Using two genotypes of Chinses cabbage, Jingcui 60 (Cd-tolerant) and 16-7 (Cd-sensitive), we characterized the root structure, subcellular distribution of Cd, CW component, and related gene expression under the Cd stress. Cysteine (Cys) supplementation led to a reduction in the Cd concentration in the shoots of Jingcui 60 and 16-7 by 65.09 % and 64.03 %, respectively. Addition of Cys alleviated leaf chlorosis in both cultivars by increasing Cd chelation in the root CW and reducing its distribution in the cytoplasm and organelles. We further demonstrated that Cys supplementation mediated the downregulation of PMEI1 expression and improving the activity of pectin methyl-esterase (PME) by 17.98 % and 25.52 % in both cultivars, respectively, compared to the Cd treatment, resulting in an approximate 12.00 %-14.70 % increase in Cd retention in pectin. In contrast, threonine (Thr) application did not significantly alter Cd distribution in the shoots of either cultivar. Taken together, our results suggest that Cys application reduces Cd root-to-shoot translocation by increasing Cd sequestration in the root CW through the downregulation of pectin methyl-esterification.

In the Karst Critical Zone (KCZ), mining and urbanization activities produce multiple pollutants, posing a threat to the vital groundwater and surface water resources essential for drinking and irrigation. Despite their importance, the interactions between these pollutants in the intricate hydrology and land use of the KCZ remain poorly understood. In this study, we unraveled the transformation mechanisms and sources of nitrogen, sulfate, and carbon using multiple isotopes and the MixSIAR model, following hydrology and surface analyses conducted in spatial modelling with ArcGIS. Our results revealed frequent exchange between groundwater and surface water, as evidenced by the analysis of δD-H2O and δ18O-H2O. Nitrification predominantly occurred in surface water, although denitrification also made a minor contribution. Inorganic nitrogen in both groundwater and surface water primarily originated from soil nitrogen (48 % and 49 %, respectively). Sewage and manure were secondary sources of inorganic nitrogen in surface water, accounting for 41 % in urban and 38 % in mining areas. Notably, inorganic sulfur oxidation displayed significant spatial disparities between urban and mining areas, rendering groundwater more susceptible to sulfur pollution compared to surface water. The frequent interchange between groundwater and surface water posed a higher pollution risk to groundwater. Furthermore, the primary sources of CO2 and HCO3- in both groundwater and surface water were water­carbonate reactions and soil respiration. Sulfide oxidation was found to enhance carbonate dissolution, leading to increased CO2 release from carbonate dissolution in the KCZ. These findings enhance our understanding of the transformation mechanisms and interactions of nitrogen, sulfur, and carbon in groundwater and surface water. This knowledge is invaluable for accurately controlling and treating water pollution in the KCZ.

Typical sources of nitrate pollution in the fragile ecological environment of karst areas, such as agricultural production activities and domestic sewage, have long attracted serious concern. However, coal development can play an equally significant role in releasing the nitrogen fixed in coal into surface watersheds in the form of nitrate, nitrite, or ammonia, consequently threatening the water quality of surface water systems in mining areas. In this study, a typical karst surface watershed system affected by coal mining activities was selected for an in-depth investigation with the aim of realistically assessing the potential contribution of coal mining to nitrogen pollution. The results reveal increasingly concerning nitrate pollution from August 2020 to November 2021 in the Huatan River watershed under the influence of anthropogenic activities, especially mining development and agricultural production. Given that the nitrogen and oxygen isotope compositions of nitrate do not support the presence of denitrification, the variation in the NO3-/Cl- ratio and the relatively stable Cl- concentration may be a reflection of nitrification. Although the leaching of atmospheric precipitation on the strata in the basin promoted the release of nitrogen associated with coal mining, the higher rate of nitrogen cycling in the oligotrophic mine water environment limited the contribution of coal mining to nitrogen pollution in the surface watershed. Specifically, the contribution of coal mining activities to nitrogen pollution in surface karst river is mainly NH4+-N, which contributes 10% or less to the nitrate input to the waters of the Huatan River. The findings thus highlight the necessity of further uncovering the geochemical cycling process of nitrogen during the transport of mine water in the coal mining environment.

Heavy metal pollution caused by acid mine drainage (AMD) is a global environmental concern. The processes of migration and transformation of heavy metals carried by AMD are more complicated in karst areas where carbonate rocks are widely distributed. Water, suspended particulate matter (SPM), and sediments are the crucial media in which heavy metals migrate and it is important to elucidate the geochemical behavior of AMD heavy metals in these environments. This study tracked AMD heavy metals from release to migration and transformation in a natural river system in a karst mining area. AMD directly impacted the hydrochemical composition of the karst water environment, but the carbonate rock naturally neutralized the acidity of the AMD. AMD heavy metal concentrations decreased gradually after the tributaries from the mining area entered the main river, with the metals tending to accumulate in SPM and sediments. The forms in which heavy metals were present were influenced by pH and their relative concentrations. Raman spectroscopy and transmission electron microscopy of sediments from the mining area suggested that the presence of an iron phase plays an important role in the fate of AMD-derived heavy metals. It is, therefore, necessary to elucidate the mechanisms of iron phase precipitation from sediments in order to control AMD-derived heavy metals in karst mining areas. This study improves our understanding of the geochemical behavior of heavy metals in karst environments and provides direction for the prevention and control of AMD in affected areas.

As important place for water storage and supply, drinking-water reservoirs in karst mountain areas play a key role in ensuring human well-being, and its water quality safety has attracted much attention. Source apportionment and ecological risks of heavy metal(loid)s in sediments of drinking-water reservoir are important for water security, public health, and regional water resources management, especially in karst mountain areas where water resources are scarce. To expound the accumulation, potential ecological risks, and sources of heavy metal(loid)s in a drinking-water reservoir in Northwest Guizhou, China, the surface sediments were collected and analyzed based on the combined use of the geo-accumulation index (Igeo), sequential extraction (BCR), ratios of secondary phase and primary phase (RSP), risk assessment code (RAC), modified potential ecological risk index (MRI), as well as the positive matrix factorization methods. The results indicated that the accumulation of Cd in sediments was obvious, with approximately 61.9% of the samples showing moderate to high accumulation levels, followed by Pb, Cu, Ni, and Zn, whereas the As and Cr were at low levels. A large proportion of BCR-extracted acid extractable and reducible fraction were found in Cd (72.5%) and Pb (40.3%), suggesting high bioavailability. The combined results of RSP, RAC, and MRI showed that Cd was the major pollutant in sediments with high potential ecological risk, while the risk of other elements was low. Source apportionment results of heavy metal(loid)s indicated that Cd (75.76%) and Zn (23.1%) mainly originated from agricultural activities; As (69.82%), Cr (50.05%), Cu (33.47%), and Ni (31.87%) were associated with domestic sources related to residents' lives; Cu (52.36%), Ni (44.57%), Cr (34.33%), As (26.51%), Pb (24.77%), and Zn (23.80%) primarily came from natural geological sources; and Pb (47.56%), Zn (22.46%) and Cr (13.92%) might be introduced by mixed sources of traffic and domestic. The contribution ratios of the four sources were 18.41%, 36.67%, 29.48%, and 15.44%, respectively. Overall, priority control factors for pollution in relation to agricultural sources included Cd, while domestic sources are primarily associated with As. It is crucial to place special emphasis on the impacts of human activities when formulating pollution prevention and control measures. The results of this study can provide valuable reference and insights for water resources management and pollution prevention and control strategies in karst mountainous areas.

Rare earth elements (REEs) have attracted much attention in recent decades due to their growing applications in high-tech industries. Coal and acid mine drainage (AMD) are considered promising alternative sources due to their high concentrations of REEs. Here, AMD with anomalous REEs concentrations was reported in a coal-mine area in northern Guizhou, China. The AMD had a total concentration as high as 22.3 mg/l, suggesting that regional coal seams may be enriched with REEs. Five segments from borehole samples, which contained coal, rocks from the roof and floor of the coal seam were collected from the coal mine site to investigate the abundance, enrichment, and occurrence of REE-bearing minerals. Elemental analysis showed that the REE contents in the coal, mudstone and limestone from the coal seam roof, and claystone from the floor (all dating to the late Permian) varied greatly, with averages of 388, 549, 60.1 mg/kg and 2030 mg/kg, respectively. Encouragingly, the REEs content in the claystone is over an order of magnitude higher than the average content reported in most other coal-based materials. The enrichment of REEs resources in regional coal seams is particularly associated with the contribution of REEs in the claystone that comprises the coal seam floor, rather than just the coal, as considered in previous studies. The minerals in these claystone samples were dominated by kaolinite, pyrite, quartz and anatase. Two types of REE-bearing minerals, bastnaesite and monazite, were detected in the claystone samples by SEM-EDS analysis, and they were found to be adsorbed by a large amount of clay minerals, mainly kaolinite. Additionally, the results of chemical sequential extraction also confirmed that the majority of the REEs in the claystone samples are mainly in their ion-exchangeable, metal oxide and acid-soluble forms, which are viable prospects for REE extraction. Therefore, the anomalous concentrations of REEs and most of them are in extractable phases, which demonstrates that the claystone from the floor of the late Permian coal seam should be a potential secondary source of REEs. Future studies will further consider the extraction model and the economic benefits of REEs from the floor claystone samples.

The present study investigates heavy metal pollution and its sources in cultivated soils in Bijie City, Guizhou Province, China. The ground accumulation index method was used to evaluate the associated risks, while correlation, principal component, and positive matrix factor model analyses were used to identify sources. The results show that the overall contamination levels, except for Cd, were not serious. Agricultural materials, industrial activities, transportation, coal combustion and atmospheric deposition, parent rock, and irrigation accounted for 19.66%, 14.11%, 14.54%, 16.33%, 20.70%, and 14.67% of the total accumulation of metals, respectively. Copper, Ni, Zn, and Cr came mainly from parent rocks; Pb was mainly from traffic emissions; Hg was mainly from coal deposition; As was mainly from irrigation; and Cd was mainly from industrial activities. The main sources of soil metals were irrigation, agricultural activities, and coal deposition in the east and industrial activities and soil-forming parent rocks in the west. The freezability difference between donkey ejaculates is a limiting factor of sperm cryopreservation. Our recent study shows that the freezability of donkey semen is related to the seminal plasma proteome. In this study, we aimed to identify the different abundance sperm proteins in good freezability ejaculates (GFEs) and poor freezability ejaculates (PFEs) using a Tandem Mass Tag (TMT) peptide labeling coupled with LC-MS/MS approach. A total of 2682 proteins were identified, among which 58 were significantly up-regulated in GFEs and 16 were down-regulated compared with PFEs. Bioinformatic analysis results revealed that the majority of different abundance proteins (DAPs) participated in copper and calcium binding, regulation of RNA biosynthetic process, positive regulation of innate immune response, and negative regulation of programmed cell death. KEGG pathway enrichment analysis showed the up-regulated proteins in GF group were mainly involved in N-Glycan biosynthesis and protein processing in endoplasmic reticulum. Our study was the first to analyze the proteome of sperm from donkey ejaculates with different freezabilities. The identified candidate proteins might be used to explore the molecular mechanism related to donkey sperm cryotolerance and might improve the screening of jacks with good sperm freezability. SIGNIFICANCE: Cryopreserved semen has been widely used in assisted reproductive technology. However, semen cryopreservation is a damaging process, which can cause oxidative stress, reduce sperm motility and motility. There are differences in sperm freezability reported to exist between or within breeds, and even between fractions coming from the same ejaculate. The freezability difference between donkey ejaculates is a limiting factor of sperm cryopreservation. The mechanisms that affect the freezing difference in sperm quality remain to be investigated, and freezability differences was found to be related to protein composition of spermatozoa. Some protein markers that can indicate good freezability or poor freezability semen have been identified in mammals. Until now, there is no information about the relationship between donkey spermatozoa proteome and freezability. Additional novel biomarkers of semen freezability in donkey spermatozoa are also needed. The identified candidate proteins might be used to explore the molecular mechanism related to donkey sperm cryotolerance and might improve the screening of jacks with good sperm freezability.

Mining activities change the groundwater level and flow conditions through pumping and drainage, which enhances the interaction between groundwater and aquifer rocks; mine drainage is discharged into the surface water system, which affects the whole karst water hydrogeochemical process. Based on hydrogeochemistry and the δ34S isotope, the hydrogeochemical processes, characteristics, and main controlling factors for waste water, karst groundwater, and surface water in a typical Carlin gold mining area and its surrounding areas were revealed. The results showed that:chemical compositions of groundwater and surface water unaffected by gold mining activities were mainly controlled by the weathering of limestone and dolomitic limestone; Ca2+, Mg2+, and HCO3- were main ions; and the water chemical types were Ca-HCO3. The mine wastewater and its downstream receiving water were affected by the dissolution of carbonate and silicate minerals, and cation exchange also played a role; the main ions were Ca2+, Mg2+, Na+, and SO42-, and the hydrochemical type gradually evolved from Ca-HCO3 to Ca-SO4. SO42- was the characteristic component in various water bodies affected by mining, and the concentration of SO42- gradually decreased from top to bottom in the well. The values of δ34S for unaffected groundwater and surface water were positive, and SO42- was mainly derived from realgar oxidation. Conversely, mine wastewater and downstream water were negative, SO42- was mainly influenced by the mixing action of realgar oxidation and meteoric precipitation, and pyrite also contributed to a certain extent. At the same time, NO3- came from agricultural fertilizer and rural domestic sewage discharge directly. Principal component analysis (PCA) further demonstrated:sulfide mineral oxidation and mining activities were the main controlling factors for the water chemical composition of mine wastewater and downstream water, whereas unaffected groundwater and surface water were mainly influenced by water-rock (carbonate rock) interactions. Agricultural fertilizer and rural sewage discharge also had a certain influence. Therefore, the study area should strengthen the interception of surface water, control-block-management of sulfide oxidation, rural domestic sewage treatment, and agricultural fertilizer.

Maternal-to-filial nutrition transfer is central to grain development and yield. nitrate transporter 1/peptide transporter (NRT1-PTR)-type transporters typically transport nitrate, peptides, and ions. Here, we report the identification of a maize (Zea mays) NRT1-PTR-type transporter that transports sucrose and glucose. The activity of this sugar transporter, named Sucrose and Glucose Carrier 1 (SUGCAR1), was systematically verified by tracer-labeled sugar uptake and serial electrophysiological studies including two-electrode voltage-clamp, non-invasive microelectrode ion flux estimation assays in Xenopus laevis oocytes and patch clamping in HEK293T cells. ZmSUGCAR1 is specifically expressed in the basal endosperm transfer layer and loss-of-function mutation of ZmSUGCAR1 caused significantly decreased sucrose and glucose contents and subsequent shrinkage of maize kernels. Notably, the ZmSUGCAR1 orthologs SbSUGCAR1 (from Sorghum bicolor) and TaSUGCAR1 (from Triticum aestivum) displayed similar sugar transport activities in oocytes, supporting the functional conservation of SUGCAR1 in closely related cereal species. Thus, the discovery of ZmSUGCAR1 uncovers a type of sugar transporter essential for grain development and opens potential avenues for genetic improvement of seed-filling and yield in maize and other grain crops.

Nitrogen (N) deficiency is a primary limiting factor for crop production worldwide. Previously, we reported root system architectural modifications of hydroponically cultured foxtail millet [Setaria italica (L.) Beauv.] to facilitate N translocation under N limitation. Here, we investigated foxtail millet for its shoot adaptation to low N in terms of internal N regulation under hydroponic culture. The results of this study revealed that the shoot N and nitrate (NO3-) concentrations significantly declined as compared to control (CK); however, the shoot over-accumulated ammonium (NH4+) under low N (LN). N shortage resulted in down-regulation of expressions of SiPetA, SiccsA, SipsbA, SirpoB, SipsaA, SiatpA, Sirps16, and SiPEPC which, undermined chloroplast functioning and CO2 assimilation for the provision of carbon skeleton. Carbon deficiency and lower activities of GS decelerated ammonia assimilation and led to over-accumulation of NH4+ in the LN-shoot, as indicated by lower concentrations of total amino acids. Thus, enhanced GOGAT activity was to assimilate NH4+ while, those of catalase (CAT), superoxide dismutase (SOD) and peroxidase (POD) were to scavenge reactive oxygen species (ROS) of NH4+ toxicity framework. The weakened chloroplast factory eventually minimized photosynthesis and reduced dry mass of the LN shoot. Such regulation of N by the shoot, perhaps, resurrected physiological functions which maintained internal mineral status under nitrogen limitation in foxtail millet.

In this study, topsoil and soil profiles were collected from typical karst areas in central Guizhou to investigate the accumulation and bioavailability of As in cultivated soils. The parent material of the investigated soil is carbonate rock, but there is strong heterogeneity in the As concentrations of soils developed in the Permian and Triassic, with average concentrations of 18.31 and 40.35 mg/kg, respectively. Additionally, the average As concentration of dolomite of the Anshun Formation (46.23 mg/kg) is slightly higher than that of the limestone in the Daye Formation (31.96 mg/kg) from the Triassic. Arsenic in the soil profiles of Triassic is also higher than that of the Permian and shows deep enrichment characteristics. Approximately 80% of the As exists as stable crystalline hydrous oxide-bound As and residual As fractions, and the bioavailability is very low.

Magnesium (Mg) is an essential nutrient for a wide array of fundamental physiological and biochemical processes in plants. It largely involves chlorophyll synthesis, production, transportation, and utilization of photoassimilates, enzyme activation, and protein synthesis. As a multifaceted result of the introduction of high-yielding fertilizer-responsive cultivars, intensive cropping without replenishment of Mg, soil acidification, and exchangeable Mg (Ex-Mg) leaching, Mg has become a limiting nutrient for optimum crop production. However, little literature is available to better understand distinct responses of plants to Mg deficiency, the geographical distribution of soil Ex-Mg, and the degree of Mg deficiency. Here, we summarize the current state of knowledge of key plant responses to Mg availability and, as far as possible, highlight spatial Mg distribution and the magnitude of Mg deficiency in different cultivated regions of the world with a special focus on China. In particular, ~55% of arable lands in China are revealed Mg-deficient (< 120 mg kg-1 soil Ex-Mg), and Mg deficiency literally becomes increasingly severe from northern (227-488 mg kg-1) to southern (32-89 mg kg-1) China. Mg deficiency primarily traced back to higher depletion of soil Ex-Mg by fruits, vegetables, sugarcane, tubers, tea, and tobacco cultivated in tropical and subtropical climate zones. Further, each unit decline in soil pH from neutral reduced ~2-fold soil Ex-Mg. This article underscores the physiological importance of Mg, potential risks associated with Mg deficiency, and accordingly, to optimize fertilization strategies for higher crop productivity and better quality.

Crop plants experience various abiotic stresses that reduce yield and quality. Although several adaptative physiological and defense responses to single stress have been identified, the behavior and mechanisms of plant response to multiple stresses remain underexamined. Herein, we determined that the leaf and vascular changes in Cucumis sativus Irregular Vasculature Patterning (CsIVP)-RNAi cucumber plants can enhance resistance to nitrogen deficiency and high-temperature stress. CsIVP negatively regulated high nitrate affinity transporters (NRT2.1, NRT2.5) and reallocation transporters (NRT1.7, NRT1.9, NRT1.12) under low nitrogen stress. Furthermore, CsIVP-RNAi plants have high survival rate with low heat injury level under high-temperature condition. Several key high-temperature regulators, including Hsfs, Hsps, DREB2C, MBF1b and WRKY33 have significant expression in CsIVP-RNAi plants. CsIVP negatively mediated high-temperature responses by physically interacting with CsDREB2C. Altogether, these results indicated that CsIVP integrates innate programming of plant development, nutrient transport and high-temperature resistance, providing a potentially valuable target for breeding nutrient-efficient and heat-resistant crops.

Reservoir sediment contamination with heavy metals produced by mining activities has aroused widespread global concern owing to its potential threat to human health. In this study, the total concentrations and speciation of heavy metals (Cd, Pb, Zn) in the Lexi (LX) and Maoshui (MS) reservoirs around the historical artisanal zinc smelting area in Southwest China were determined, and pollution indices were applied to assess the pollution levels and potential ecological risks of the two reservoirs. The results showed that all the detected samples in the two reservoirs presented significant metal accumulation, especially for Cd, as compared with the soil background values in Guizhou Province. Between the two reservoirs, the vertical distribution characteristics of each metal in sediment columns were similar. The heavy metal concentrations of the three columns in the LX reservoir reached their maxima at 35, 15, and 10 cm and showed a trend of first increasing and then decreasing overall. However, the heavy metal contents of the three columns in the MS reservoir all exhibited wave-like characteristics in the vertical direction, and all of them reached a relatively obvious high point at approximately 5 and 30 cm. The geoaccumulation index (Igeo) and potential ecological risk index (RI) indicated that Cd was strongly enriched and represented the main risk factor, and the pollution level of the MS reservoir was significantly higher than that of the LX reservoir. Furthermore, the effect coefficients (ERMQ) confirmed that the two reservoirs are likely to have toxic impacts on aquatic organisms and need to be controlled and mitigated. The speciation analysis of heavy metals revealed that Cd was primarily in the acid-extractable fraction (69.57%, 68.28%), Pb was chiefly in the reducible fraction (55.24%, 42.18%) and oxidizable fraction (22.60%, 38.02%), and Zn was mainly in the oxidizable fraction (32.54%, 37.65%) in the LX and MS reservoirs, respectively. The ratios of the secondary phase and primary phase (RSP) and risk assessment code (RAC) evaluation demonstrated that Cd in the sediments of the two reservoirs presents a very high potential ecological risk, and Pb and Zn were at medium to high ecological risk levels. This study highlighted that the artisanal zinc smelting activities had caused serious heavy metal pollution in reservoir sediments, posing a threat to the local ecological environment.

Coal resources are widely distributed in Guizhou province, China and environmental pollution caused by coal mining is becoming increasingly serious, especially mine drainage. A total of 120 mine water samples collected from different coalfields were analyzed to investigate the hydrogeochemical characteristics and assess the water quality for drinking, domestic, and irrigative purposes. Water samples had a pH of 1.90-9.12 and most of them were acidic or weakly acidic. Total dissolved solids (TDS) ranged from 254 to 13,944 mg/L and correlated closely with the electrical conductivity (EC). The coal mine drainage of Guizhou was characterized typically by low pH value, high Fe and SO42- concentration, which were mainly attributed to oxidative weathering of pyrite. The most dominant type for abandoned coal-mine drainage was Ca-Mg-SO4, while that of the underground drainage for active mine were mainly Na-SO4 and Na-HCO3 because of high concentration of Na+ from the dissolution of evaporites and clastic rocks. High concentration of the TDS, SO42-, Fe, Mn, et al. made it unsuitable for drinking and domestic use, but part of the coal mine drainage could be used for irrigation at some sites.

Variation in donkey sperm freezability (capacity to withstand freeze-thawing) between ejaculates is a limitation for sperm cryopreservation. Seminal plasma proteins are essential for sperm function and also related to individual differences in sperm freezability. A Tandem Mass Tag (TMT) peptide labeling combine with a LC-MS/MS approach was conducted to quantitatively identify the seminal plasma proteins differentially abundant in ejaculates with optimal freezability characteristics (GFE) compared with those with suboptimal freezability characteristics (PFE). A total of 866 proteins were identified, and 99 ejaculates were in larger abundance in GFE samples. Differentially abundant proteins (DAPs) were subjected to intensive bioinformatic analysis. The majority of DAPs were involved in metabolic processes, oxidation-reduction processes and biological regulation. Results from functional protein analysis suggested that proteins functioned in oxidoreductase activity and acid phosphatase activity. This is the first report where there were analyses of the proteome of seminal plasma from donkey ejaculates with different freezability and to identify candidate proteins that could be used to explore the molecular mechanism related to donkey sperm cryotolerance. In this study, there also was elucidation of biomarkers for the early identification and selection of donkeys with optimal semen freezability.

This study aimed to assess the sexual behavior of jacks mounting jennies in estrus and determine the daily sperm output (DSO) and spermatogenic efficiency using two equations to calculate testicular volume (TV). Eight sexually rested mature jacks, aging 5 to 10 years old, had semen collected once a day for 10 consecutive days using jennies in good standing estrus for mounting. Sexual behavior and semen parameters were assessed during each collection. Testicular measurements of height, width, and length were taken immediately before the first semen collection, and these measurements were used to calculate TV. After that, the TV was used to predict the DSO. The average total sperm number (TSN) obtained on days 8 to 10 was deemed the actual DSO. Differences in the predicted vs. the actual DSO were used to calculate the spermatogenic efficiency. In addition, the actual DSO was also used to calculate the number of inseminating doses a jack could produce for both on- and off-site breeding. Jack's sexual behavior and sperm motility did not vary across collection days. Sperm concentration and TSN reduced over time (P < .05). The actual DSO was 9.1 ± 4.1 billion, and the predicted DSO varied from 4.7 to 18 billion. Spermatogenic efficiency varied from 50 to 150% based on jack and the equation used to calculate TV. The number of inseminating doses ranged from 15 to 47 at 300-500 million progressively motile sperm (pms)/dose for on-site breeding. In contrast, the number of breeding doses with cooled-shipped semen (1 billion pms/dose) varied from 4 to 14 doses across donkeys. In conclusion, sexual behavior was not affected by daily semen collections. Sexual rest did not affect sperm motility. The predicted DSO varied with the equation used to determine TV. Clinically normal donkeys have high spermatogenic efficiency, which confirms previous histology reports.
